The complete molecular entity comprises a 37-amino-acid amylin-analogue backbone carrying an intramolecular disulfide bridge between its two cysteine residues (the native-amylin-equivalent bridge; backbone positions 2 and 7), a C-terminal amide, and an eicosanedioic acid (C20 fatty diacid) attached through a gamma-glutamyl linker to the N-terminal-region lysine. The lipidation confers albumin binding and the protracted pharmacokinetic profile engineered in its development (Kruse et al., 2021).
